Lightsheet Z.1 Imaging Workshops for Cleared Samples

The event is organised jointly by Dr Katherine Lau from ZEISS, Dr Chris Lagerholm from the Weatherall Institute of Molecular Medicine and Dr Andrew Jefferson from the Micron Advanced Bioimaging Unit at the University of Oxford. Dr Jacques Paysan, Product and Application Sales Specialist from Carl Zeiss Microscopy GmbH will be on hand during the imaging workshops as the demonstrator.

ZEISS Lightsheet Z.1:
The ZEISS Lightsheet Z.1 microscope is excellent for imaging cleared samples. Light Sheet Fluorescence Microscopy (LSFM) uses a thin plane of light (light sheet) to optically section transparent tissues or whole organisms that have been fluorescently labelled. Using a light sheet, and illuminating only the image plane during acquisition, lightsheet imaging is extremely gentle and induces almost no photobleaching.

The emitted light is collected by a separate objective positioned perpendicular to the light sheet plane, allowing the imaging of the (excited) tissue during the illumination of only a single thin section of the sample. This enables submicron resolution images to be rapidly acquired from larger samples (in centimetre range), such as mouse brain, tumours and spheroids. The orientation of the sample in Lightsheet Z.1 allows sample to be rotated 360 degrees and enables Multiview experiments, i.e. for Z-stack to be acquired from multiple angles, thus increases the image quality and resolution.

Workshop Details:
The Lightsheet Z.1 microscope is being upgraded to clearing ready (compatible with cleared samples, at refractive index of 1.45 +/- 0.03). We would like to invite you to join us at the ZEISS Lightsheet Z.1 imaging workshop for cleared samples at the Micron Imaging Facility at the University of Oxford to experience this first hand.

There are various demo slots available throughout the three day period. Each slot can accommodate a maximum of four people. Samples should be cleared with a non-solvent based clearing medium at RI 1.45 +/- 0.03, e.g. CLARITY, TDE, LUMOS and FocusClear.
